I don't know St Petersburg. It may be that there is no public transport on that route, but that seems unlikely if it is in the city itself. If you've set your location as said by catymag, have you got all transit types enabled as Giomen suggested? Tools>Settings>Navigiation>Transit Types:
@Neil, I think this somehow cant work like it should. [Only registered and activated users can see links. Click Here To Register...]

Yes, i agree. But he stated in [Only registered and activated users can see links. Click Here To Register...] that he has the full CX's for Moscow & St Petersburg only in his unit. I didn't see that pic before, is that his screenshot from somewhere? So I did assume there is no other older CX or a CN Russia of any version on his unit to conflict. If he has, that will be it Caty and you're very perceptive as usual. ;)

TA-DA!!! Public transportation works now. The problem was: old and new CX maps in the unit at the same time. Once I removed old CX maps, new CX maps began calculating public transportation routes. Somehow old and new CX maps conflict with each other, even though old CX maps were unchecked on the Map Info screen. CX maps do not seem to conflict with regular NT maps covering same area.

Nice :)
Always remove old maps (backup first on pc) from Nuvi if you have a newer version. City Navigators, CXs or Topos.
Nuvi often wouldn't show ether new nor old ones or would give weird results.
Deselect also maps covering the same area (except small cx), choose one and deselect the others when not using them. You'll avoid some problems, not always noticeable at once or predictable.
